Study on Radioimmunoassay for Human Chorionic Gonadotropin in Serum of Normal Pregnant Women

Abstract

|
It is not practical to quantitate HCG in the serum of normal pregnant women by utilizing hemagglutination inhibition technique and bioassay. For the practical application of radioimmunoassay and technical familiarization, quantitative determination of HCG were performed by means of radioimmunoassay-Antisera were developed against human chorionic gonadotropin. Highly purified HCG was radioiodinated with Na-125I by the method of Greenwood, Hunter and Glover and labeled HCG was separated by Sephadex G-100 column chromatography. Separation cf antibody-bound from ¢¥free HCG-125I was accomplished by a double antibody technique and the obtained results are as following.
1) The sensitivity of radioimmunoassay in this study could detect 10 mIU/ml of. HCG.
2) The quantitative determination of HCG in the serum of 137 pregnant women at various gestational period revealed that HCG has been sharply increased after 5th week of gestation and reached to the peak in 9-12th week, giving an average of 106, 000 IU/L, then gradually declined to reach a point of minimum-levels in 20th week and 20, 000-30, 000 IU/L has been maintained until the end of pregnancy.
3) Comparison between HCG levels in serum of pregnant women at various gestational period and that of urine determined by HAIR revealed very similar changing pattern.
